Abstract In order to treat bacterial infections successfully in the future, the mechanism of action of metallo‐β‐lactamases (MβLs) needs to be fully understood so that clinically useful inhibitors and new therapeutic approaches can be developed. This microreview describes recent developments and the achievements towards the design of MβL biomimetics to aid in the mechanistic and structural understanding of these enzyme systems. We highlight the known inhibitors and substrates employed in these studies, the different ligands and complexes employed, and the experimental and computational studies undertaken to explore the possible mechanisms of action of the MβLs.
