The Na + ‐dependent binding of γ‐aminobutyric acid (GABA) in rat cerebral hemispheres was studied by subcellular fractionation and double‐labelling procedures. The distributions of tracer amounts of [2,3‐ 3 H] GABA and [U‐ 14 C] surcrose acquired by P 1 (crude nuclear) and P 2 (synaptosomal, mitochondrial) particles during homogenization at O° C were compared in the presence of 40 m M Na + and in the absence of added Na + . Results indicated that addition of Na + to the homogenizing fluid promotes extra binding of GABA to both P 1 and P 2 particles and that these processes are age‐dependent, but different. GABA binding to P 1 particles increased with age until 21‐22 days after which it remained rather constant. On the other hand, a more dramatic developmental change occurred in the binding of GABA to P 2 fractions (which contain the majority of the nerve endings) which was maximal in the brains of rats 11 and 15 days old and which declined in older rats.
