Studies of central cholinergic mechanisms, cholinergic drugs, and Aizheimer's disease (AD) have been reviewed and analyzed. Although the pronounced cholinergic deficit that exists in the brains of AD patients is associated with memory loss and other cognitive dysfunctions, cholinergic “replacement therapy” is of limited benefit in trating AD. Recent findings concerning the roles of nerve growth factor, “excitotoxic” amino acids, and amyloid deposition in AD migh lead to mroe fruitful therapeutic approaches.
