Proteinase‐activated receptor (PAR) 1 and PAR 2 are abundantly expressed in the gastric mucosa and smooth muscle. PAR 2 modulates a variety of gastric functions, mediating secretion of mucus and pepsinogen, suppression of acid output, an increase in mucosal blood flow, and smooth muscle contraction/relaxation. PAR 2 is considered primarily protective in the gastric mucosa. PARs other than PAR 2 might also be involved in the modulation of gastric functions. PARs, especially PAR 2 , would thus appear to be novel targets for the development of drugs for treatment of gastric mucosal diseases. Drug Dev. Res. 60:9–13, 2003. © 2003 Wiley‐Liss, Inc.
