Effect of colestipol on gastrointestinal absorption of chlorothiazide in man

The effect of colestipol on gastrointestinal absorption of chlorothiazide was studied in 10 adult patients. A 58% decrease in 24 hour cumulative urinary excretion of chiorothiazide was observed when the drug was ingested simultaneously with colestipol. A decrease of 54% was observed when the two drugs were administered one hour apart. The in vitro binding of chlorothiazide to colestipol is pH‐dependent, with 64% of the drug bound to the resin at pH 6.9 as opposed to 9% bound at pH 1.0. This implies that the in vivo binding of the drug takes place primarily in the small intestine.
