Absorption of intestinal antiseptics derived from 8‐hydroxyquinolines

Urinary excretion for 10 hours after a small oral dose of several halogenated 8‐hydroxyquinolines was spectrophotometrically determined in a series of normal human sub;ects. No free forms of drug were found. The determinations were made on the glucuronic acid con;ugates, and the excretions in per cent of the dose were: di‐iodo‐8‐hydroxyquinoline, 4.6 per cent; di‐bromo‐8‐hydroxyquinoline, 10.2 per cent; chloro‐iodo‐8‐hydroxyquinoline, 12.6 per cent; di‐chloro‐methyl‐8‐hydroxyquinoline, 34.9 per cent. Three normal children and two patients (suspected of having 8‐hydroxyquinoline toxicity) who received chloro‐iodo‐8‐hydroxyquinoline had a similar excretion. Human absorption and ocular toxicity are discussed.
