Examination of the problems involved in the design of investigations of drugs in man indicates that there is no routine approach which provides meaningful answers in all cases. Appropriate control of the experiment in man is especially elusive. Each case has to be considered separately. It is suggested that with the increasing recognition of the need for controlled studies in man, some aspects of the problems involved have been generally recognized and better handled than previously, hut that the problems inherent in the selection of the most appropriate subject are often ignored.
