The key to unlocking the secret? The nonstructural protein NS5A has no known enzymatic function, but it is essential for hepatitis C virus (HCV) replication and has been proposed to function as a regulator for key replication events. The first clinically validated NS5A inhibitor, developed via chemical genetics screens with HCV replicons, was recently described and this work is highlighted here.
