Main observation and conclusion Ten new characteristic dihydroagarofuran sesquiterpenoids and three known ones were acquired from the Celastraceae plant Tripterygium wilfordii . Their structure characterizations were done through spectroscopic data, while compounds 10—12 were further confirmed by single crystal diffractions. Several dihydroagarofurans protected PC12 cells against H 2 O 2 ‐induced cytotoxicity as detected by CCK8 assay. Compounds 5—7 and 13 significantly increased the levels of reactive oxygen species (ROS) and superoxide dismutase (SOD), and decreased malondialdehyde (MDA) levels. In addition, compounds 5—7 and 13 activated the Nrf2 signaling pathway and increased Nrf2, NQO1, and HO1 expression levels in PC12 cells after H 2 O 2 treatment.
