Bioorthogonal fluorogenic reagents that produce after chemical activation fluorescent products represent superior probes for cellular imaging. Due to the improved signal‐to‐noise ratio, they can be applied in no‐wash fluorogenic labeling experiments. The present study reveals a set of coumarin‐tetrazine light‐up probes, which produce bright dyes in reaction with TCO or BCN dienophile‐tagged molecules and enable fast bioorthogonal labeling in live cells.
