Generation of novel responsive fluorescent sensors requires synthetically versatile fluorophore cores. An intermediate for modular synthesis of tri‐functionalised 4‐amino‐1,8‐naphthalimide fluorophores was enabling the mix and match of sensing and targeting groups as well as additional functionalities. Using this intermediate a set of hypoxia‐responsive fluorophores was prepared, including an extracellular analogue that can image the hypoxic region of a tumour spheroid.
