A new hybrid water soluble acyclic receptor that strategically merges the properties of CB[n] and Pillar[n]arene (PA[n]) is reported. This receptor is able to fabricate supramolecular vesicles for drug encapsulation and triggered release through co‐assembly with an amphiphilic guest. The cover image emphasizes the structures of the receptor and bilayer vesicles while CB[6] and PA[5] are fitted in the background to indicate the conceptional relationship between the receptor and CB[n] and PA[n].
