A new class of spiropyran‐based protease inhibitor is reported that reversibly photoswitches between an active spiropyran (SP) isomer and a less active merocyanine (MC) isomer on irradiation with UV and visible light, respectively. Switching occurs in solution and also when attached to the surface of a microstructured optical fiber (MOF). The center of the image shows a space‐filling model of the target protease, α‐chymotrypsin and a MOF is depicted in the upper‐right‐hand corner.
