A fast, high‐yielding and reliable method for the synthesis of DNA‐ and RNA 5′‐triphosphates is reported by C. Meier et al. This method allows the inexpensive preparation of 5′‐triphosphorylated oligonucleotides with efficient coupling reactions and extremely high phosphorylation efficiencies. An important advantage as compared to other methods is that the complete reaction sequence can be performed in an automated way by using a standard oligonucleotide synthesizer.
