Five base‐modified nucleoside triphosphates (dNTPs) adorned with functionalities that play a central role in organocatalysis (i.e., proline, urea and sulfamide groups) were synthesized. In addition, these dNTPs were shown to be compatible with methods of in vitro selection since they are good substrates for polymerases, both in primer extension reactions and the polymerase chain reaction (PCR).
