Activity‐based protein profiling (ABPP) is a powerful approach for profiling and bioimaging many therapeutic targets for bioactive molecules in situ. The cover feature picture shows an activity‐based probe for Bcl‐2/Mcl‐1 profiling and visualizing. Integrating the Bcl‐2/Mcl‐1 binding ligand, fluorescent reporter group and photoreactive group into the same scaffold creates a multifunctional probe that can label, capture, enrich, and bioimage Bcl‐2/Mcl‐1 proteins both in vitro and in cells.
