Tracing aspartic proteases: We present the design of activity‐based probes for cathepsins D that are effective in a complex proteome. The probes consist of a binding module with a statin‐containing peptidic core, a warhead module bearing a photoreactive group, and a reporter tag. The fluorescent and biotin tags make the probes a powerful tool for a wide range of “in‐gel” and “on‐blot” proteomic applications.
