Extra‐mild nondenaturing protocol: Fishing for unaltered protein in a lysate is a prerequisite for identifying and analyzing the architecture of a protein complex. We have shown that a noncrosslinked protein–ligand complex can resist bio‐orthogonal Staudinger ligation. The resulting target–probe–label entity is compatible with nondenaturing protein analysis.
