Sequence‐specific triplex formation significantly accelerated PN bond digestion of triplex‐forming oligonucleotides involving a P3′→N5′ phosphoramidate linkage. By incorporation of a fluorophore and a quencher into the triplex‐forming oligonucleotide probe, sequence‐specific double‐stranded DNA sensing was successfully achieved.
