High‐Throughput‐Compatible Assay for glmS Riboswitch Metabolite Dependence

A fluorescent varient of the glmS ribozyme was used to develop a fluorescence polarization‐dependent screeening and metabolite‐detection assay. The metabolite‐induced in‐cis cleavage of the naturally occurring ribozyme could be shown to be stable and allows efficient monitoring of the ribozyme‐catalyzed reaction even in bacterial extracts. The standard deviations and z ′ values of the assay format indicate the high‐throughput screening compatibility of the applied system for the identification of compounds that act on RNA structures.
