Not only the first! We show that DNA polymerases inspect mismatches within the primer–template complex up to four positions distal to the catalytic center, as shown. Insights into enzyme transitions were drawn by chemical probes and extended to direct analysis of single nucleotide variations through allele‐specific PCR.
