Detective work . To detect a specific mRNA, a 5′‐DNA‐RNA‐DNA‐3′ template has been designed and immobilized on solid phase, this allows polymerase labeling and extension immediately following RNase H digestion. After alkaline phosphatase labeling and substrate addition, a chemiluminescence single can be detected as per the scheme.
