Optimization of the experimental procedures in fast atom bombardment (FAB) mass spectrometry of protected and free peptides of up to m / z = 2000 u with a quadrupole gas chromatograph/mass spectrometer and a Capillaritron direct insertion probe gun tip has been carried out. The influence on the molecular ion intensity, the background noise and signal to noise ratio of nine experimental parameters relating to gun, sample and data acquisition was studied. Peptides of up to m / z = 2000 u can give good FAB mass spectra with a good signal to noise ratio and an acceptable mass accuracy.
