Mass spectra and fragmentation pathways of some achiral and a number of chiral 1,4‐benzodiazepine‐2‐ones, and those of their in vitro biotransformation products are reported and discussed. Various derivatives substituted in position 3 are shown to be useful for the determination of fragmentation pathways from low resolution spectra.
