Roe et al . analyze unconstrained tetramer segments of a nanosheet‐forming peptoid 28mer, to elucidate stabilizing interactions using NMR spectroscopy. Surprisingly, the N ‐terminal tetramer was found to be conformationally stable in aqueous solution with a trans‐cis‐trans amide backbone pattern. All backbone dihedral angles fall within the expected minima of the peptoid energy landscape. Peptoid‐specific 2D and 3D NMR methods were developed to assist in resonance assignment that should be generalizable to a wide variety of sequences. (DOI: 10.1002/bip.23267 )
