The pE‐MAP technology allows for functional dissection of macromolecular assemblies by genetic interrogation of point mutants. Application to yeast RNA polymerase II has enabled functional assignment of its regions and uncovered details related to its catalytic activity. On pages 706–713 of this issue, Braberg et al. discuss the key applications of pE‐MAP, as well as its potential use for personalized medicine when extended to human cells.
