A simple and highly efficient method was developed to prepare a synthetic poly(ethylene glycol) (PEG) derivative, which can be used for conjugation to histidine residue. This derivative reacted with sodium nitrite and the intermediate 4‐diazo‐N‐mPEG‐benzamide proved to have a good performance for histidine modification of lysozyme. Then the pegylated lysozyme was separated and its bacteriolytic activities were determined. © 2010 Wiley Periodicals, Inc. J Appl Polym Sci, 2011
