The death cap toxin α‐amanitin has received increasing attention as a toxin component of antibody–drug conjugates in cancer research. A new route to this bicyclic octapeptide that is based on a convergent approach in solution phase is reported by R. D. Süssmuth and co‐workers in their Communication on page 5500. The straightforward synthesis gives access to the non‐proteinogenic amino acids l ‐6‐hydroxytryptophan and (3 R ,4 R )‐ l ‐4,5‐dihydroxyisoleucine on multigram scale.
