The light‐controlled regulation of the activity of protein subdomains is possible with a new approach termed apta‐caging, the development of which is reported by A. Heckel, G. Mayer et al. in their Communication on page 11912 ff. The aptamer HD1 is bound by a photocleavable linker to the regulatory Exosite I of thrombin. As a result, the thrombin activity can be spatially and temporally controlled by light irradiation. Owing to the large interaction area of the aptamer mask, no knowledge of the protein structure is required.
