The first X‐ray analysis of a B 12 protein , the B 12 ‐binding domain of the methionine synthase of Escherichia coli , led to a surprising finding: there the B 12 derivative, methylcobalamin, is not bound in the “base‐on” form present in solution and in the crystal. Instead a histidine residue coordinates at the cobalt center of the protein‐bound cofactor in place of the nucleotide function.
