The protected enediyne 1 is the first example of an estramycin, a new family of potential anticancer antibiotics, derived from estradiol. Important for therapeutic applications is the cyclization of the enediyne unit, which proceeds via a diradical intermediate. R 1 R 3 Ac; R 2 SiMe 3 .
