Chemoenzymatic gene synthesis in combination with cloning in E. coli enables the synthesis of larger proteins such as human gamma interferon. Chemical DNA synthesis followed by enzymatic coupling and cloning of the individual gene segments and ligation of the subfragments then yields the gene for the biologically active gamma interferon.
