Chemical investigations of the leaves of the Australian endemic rainforest plant Denhamia pittosporoides afforded two new (denhaminols I and J) and four known dihydro‐ β ‐agarofurans. The chemical structures of all natural products were elucidated by detailed analysis of spectroscopic data. The absolute configuration of denhaminol I was established by X‐ray crystallography. Several secondary metabolites were found to inhibit leucine transport in a human prostate cancer cell line with better potency than the L‐type amino acid transporter (LAT) family inhibitor, 2‐aminobicyclo[2.2.1]‐heptane‐2‐carboxylic acid.
