A paper‐based hydrophobic/lipophobic surface with high liquid drop adhesion capable of carrying out sensitive biochemical assays directly is reported. Hydrophobic sol‐gel derived fluoroalkyl‐silica is used to develop the hydrophobic/lipophobic surface, which demonstrates superior confining abilities. The reported hydrophobic/lipophobic surface could retain aggressive liquids such as organic solvents and surfactant solutions whereas the methyl‐silica based surface fails to function. Because of the superior confining abilities, this paper‐based hydrophobic/lipophobic surface is competent for sensitive heavy metal ions detection and bacteria assay where aggressive liquids are necessarily used. The condensing‐enrichment effect on the hydrophobic/lipophobic surface renders this paper‐based analytical device (PAD) high sensitivity, and the high adhesion of the surface offers the PAD robustness. Facile fabrication of such surface and ease of operation for subsequent sensing promise a great potential for the PAD.
