A reversible PEGylated prodrug integrates the advantages of PEGylation and responsiveness. J. Ding and colleagues reveal on page 844 that the prodrug exhibits acid‐accelerated drug release, improved cellular internalization and cytotoxicity in vitro, and upregulated intratumoral accumulation, antitumor efficacy, and security in vivo.
