The orthogonal genetic system is an artificially engineered system that runs in parallel with the native genetic system. In article number 2000252, Huimin Zhao, Shuobo Shi, and co‐workers describe the advances and future challenges in developing such systems. The orthogonal genetic system can be controlled at will and is widely applicable in biosynthesis and thus represents a powerful approach for synthetic biology.
