Genetically encoded chemical crosslinking (GECX) via bioreactive unnatural amino acid (Uaa) enables the capture of weak and transient protein‐protein interactions in live cells for identification with mass spectrometry. In article number 2000308, Chao Liu, Shang‐Tong Li, Kui Li, Lei Wang, Bing Yang, and co‐workers develop a new software OpenUaa to analyze Uaa‐crosslinked peptides, which uniquely considers all fragment ions for high sensitivity and identify direct protein interactome with the GECX‐OpenUaa strategy.
