PTEN protein levels are regulated through ubiquitination. Here, we identify BAP1 as a deubiquitinase that physically binds to and deubiquitinates PTEN, thereby inhibiting PTEN degradation. Stabilized PTEN suppresses prostate cancer progression by inhibiting Akt phosphorylation.
