A literature review of cytotoxic agents used in the chemotherapy of recurrent and/or metastatic adenocarcinoma of the uterus is presented. Fifteen different agents have been used in 126 patients with this gynecologic malignancy. Measurable objective responses have been reported in 16% of patients. 5‐Fluorouracil, cyclophosphamide, and chlorambucil have been most extensively evaluated. Data on other agents used in a limited number of patients are presented. In spite of the abundance of clinical data reporting the results of chemotherapeutic trials which have been published in recent years, there is a paucity of information dealing specifically with cytotoxic chemotherapy of endometrial adenocarcinoma. Recommendations for trials with cytotoxic agents, cytotoxic‐hormonal combinations, adjuvant drug studies, and immunologic approaches is presented. Optimal chemotherapy of adenocarcinoma of the endometrium is not evident. It is hoped that this review will stimulate randomized prospective clinical trials which may lead to a more optimistic future for these patients.
