Murine testicular tumors analogous histologically to human germinal testis tumors were produced according to a previously described technique. The effects on this model of actinomycin D and of a 4‐drug regimen using 5‐Fluorouracil, vincristine, methotrexate, and Cytoxan were compared. Both chemotherapeutic regimens tested produced a demonstrable effect on the incidence, diameter, and differentiation of tumor foci. The 4‐drug regimen was found to be more effective than actinomycin D. A single‐dose actinomycin D protocol was shown to be less effective than a course of injections, and the desirability of maximal drug doses was suggested. Greatest drug effects were elicited when the tumors were small and poorly differentiated.
