Effects of antitumor agents and hyperbaric oxygen in normal and tumor‐bearing rodents

Investigation of the toxic and antitumor effects of 10 chemotherapeutic agents and hyperbaric oxygen was undertaken with 5 tumors in 9 rodent species. Pentobarbital sodium greatly enhanced the toxicity of hyperbaric oxygen in mice. D‐54 naponate, a dihydrotriazine, was significantly more toxic when administered with hyperbaric oxygen; the toxicity of other drugs tested was not altered significantly by high oxygen pressure. Prolongation of survival time and inhibition of tumor growth by drug alone was enhanced by a combination of hyperbaric oxygen and OMF‐59, a cyclophosphamide congener. None of the other drugs studied showed significant increase in antitumor activity with hyperbaric oxygen. In general, the results show little enhancement of antitumor effect by hyperbaric oxygen; however, the specific techniques of oxygen administration and the types of animal, drug, and tumor tested are all critical determinants of results.
