An improved protocol for the synthesis of oligonucleotides containing a 3′−3′ phosphodiester linkage, for use in an alternate strand triple‐helix formation approach, is reported. Three 16‐mers and one 24‐mer have been efficiently synthesized and the corresponding triplexes have been analyzed by thermal denaturation experiments and CD spectroscopy.
