Optical imaging of microscopic objects provides valuable information in various fields of research ranging from physics to engineering and to medical diagnosis. Particularly in biophysics, optical microscopic techniques are invaluable methods for investigating important scientific questions. Conventionally, bright-field microscopy and fluorescent techniques have been extensively used in biophysics. Recently, holographic microscopy techniques have been exploited for the study of biophysics. Due to the level-free and quantitative imaging capability of holographic microscopy, its uses have grown rapidly. To provide an overview of how holographic microscopy can be exploited to address various biophysical problems, we summarize recent progress on holographic microscopy techniques and their applications in biophysics.
