This research aimed to know about multiplication shoot of cotton plant (Gossypium spp.) varieties Kanesia 15 using a combination of BAP and NAA by  in vitro. The research was conducted in October 2017 to January 2018 at the Laboratory of Plant Tissue Culture, State Polytechnic of Jember. The experimental design used was Factorial Completely Randomize Design (CRD) consisting of 2 factors with 4 replications. First factor was the BAP consisted of four levels: 0.00 ppm, 1.50 ppm, 3.00 ppm, and 4.50 ppm. Second factor was the NAA which consisted of three levels: 0.00 ppm, 0.10 ppm, and  0.50 ppm.  Observation data obtained were analyzed using Analysis of Variants (ANOVA, if significant (*) followed by further Duncan Multiple Range Test (DMRT) at a level of 5%.. The results showed that BAP on the media had no significant effect on emergence of shoots, number of shoots, and diameter of explants, but gave a very significant effect on explant height, number of internodes, number of roots, and root length.
