Bu calismada Turkiye’de Kayseri civarinda yetistirilen Holstayn irki sigirlarda (n=352) leptin (LEP) ve hipofiz bezine ozgu transkripsiyon faktoru -1 (Pit-I) genlerinin polimorfizmlerinin belirlenmesi amaclanmistir. Pit-I-HinfI ve LEP-Sau3AI polimorfizmlerinin belirlenmesi amaci ile polimeraz zincir reaksiyonu (PZR) ve restriksiyon parca uzunluk polimorfizm (RFLP) analizi yapilmistir. Pit-1 geni icin 600 bc’lik ve LEP geni icin 422 bc’lik uzunluk amplifiye edilmistir. Calismada, Pit-1 ve LEP geni allellerinin her ikisinde de A ve B allelleri olmak uzere iki iki tip allel gozlendi. En yuksek frekans Pit-1 geni icin A allelinde (0.87), LEP geni icin ise B allelinde (0.68) olarak belirlendi. Ki-kare test sonuclarina gore, incelenen irkta, her iki gende de Hardy-Weinberg dengesinden anlamli bir sapma gozlendi (ayni sira ile; X2:40.75, p<0.001ve X2:8.4, p<0.01)
