A staining procedure is described in which the basic dye crersyl violet is shown to possess properties which makes its use advantageous in the staining and cytophotometric estimation of ribonucleic acid. Cresyl violet is shown to stain ribonucleic acid quite specifically when used at pH 4.2, and the resulting in situ staining is shown to follow the Beer-Lambert Law over the range of thicknesses and concentrations found in microscopic sections.
