Results obtained by the PbS method and by the post-coupling method of Rutenburg and Seligman are closely similar. However, there are minor differences which appear to be due partly to the presence of more than one acid phosphatase in the tissues, partly to diffusion artifacts. Results of techniques recommended so far for the histochemical demonstration of acid phosphatase must be interpreted with caution, especially with respect to finer details.
