A study was made of the influence of experimental variables, including electrolyte balance, pH, tonicity, length of incubation and dimensions of tissue slice, on the in vitro reduction of tetrazolium. The most discriminating approach included examination of both the histochemical and quantitative expressions of the deposition of formazan. Some of the limitations of neotetrazolium (NT) and 2,3,5 triphenyl tetrazolium chloride (TTC) are discussed, NT being less suitable for quantitative estimates of reduction values.
