Available data are consistent with the interpretation that the reaction between orcein and elastin is due to the formation of hydrogen bonds. The selectivity for elastin of this reaction in acid alcohol solution is probably due to the uniquely low positive charge of elastin in such solutions. The use of alcohol solutions is necessary to solubilize the positively charged orcein fractions.
