Replication of positive-strand RNA viruses is thought to occur in tight association with remodeled host cell membranes. Paul et al. (p. 10612–10627) use a new approach to specifically isolate membranes remodeled by hepatitis C virus (HCV) and show that active viral replicase complexes are associated with double-membrane vesicles. This result suggests that double-membrane vesicles are the site of HCV replication. The approach described here might be of general use to study the molecular composition of membranous replication factories of positive-strand RNA viruses.
